Kinases play roles within cells that drive multiple biological processes. They represent a broad class of drug targets, the aberrant activity of which is involved in a range of diseases, including cancer, cardiovascular, inflammatory and fibrotic diseases.
As kinases are more structurally diverse in the inactive state, there are greater opportunities to achieve selective inhibition than in the active state. In addition to this selectivity and safety advantage, targeting inactive kinases eliminates need to compete with high intracellular levels of ATP, providing an important potency / efficacy advantage. Kinagen's drug discovery approach also affords unique mechanistic opportunities to inhibit dimerization of targeted kinases and pseudokinases that are difficult to address by other means.
